USP6 was immunoprecipitated from 1 mg of FLAG-USP6 stable inducible cell line using 5 ug of anti-USP6 [S566C]. The immunoprecipitate, along with lysate from the FLAG- USP6 stable inducible cell line (+/- tetracycline) was, subjected to SDS-PAGE, transferred to nitrocellulose membrane and immunoblotted with either anti-FLAG or anti- USP6 [S566C] antibody. Binding of the primary antibody was detected using rabbit peroxidase conjugated antisheep IgG antibody (1:10.000 dilution, Pierce) followed by enhanced chemiluminescence (ECL, Amersham).
