note: EcoRV cleaves the pHISTEV30a plasmid twice, once before the coding
sequence for the TEV protease cleavage site and once in the polylinker. Digestion of the pHISTEV30a vector with EcoRV thus removes the TEV protease cleavage site. The coding sequence for the TEV protease cleavage site was therefore included in the forward primer.
